Depends on what you mean?
As a businessman, he was at their level at one point particularly around the late 2000's but that lawsuit with Ross's BM took him down a few notches. Nonetheless, he's still making some good moves business wise in different areas.
As an artist, 50 is better than Puffy but Jay's catalog is much stronger. GRODT is better than most Jay albums but Jay has more than one critically acclaimed album unlike 50.
As a rapper, he's way better than Puffy who's hardly a rapper to begin with but not close to Jay's level. 50 is more charismatic than he is lyrical.
Popularity wise? He's bigger than both of them overseas. America is debatable though. I think more Americans know 50 than Puff but Jay is more known than both cause of his connections with various business ventures, Beyonce, Kanye, and his sheer longevity as an artist who's been in the game since the late 80's and is still able to catch a buzz today.
So, in essence, it really depends on what you mean.
