There are numerous reasons we cannot physically survive the trip to mars safely.
I discussed some of them in JBO.

When you are in space for over 14 days your body begins to shed its skeleton to adapt to weightlessness.
This is the reason they need to have artificial gravity machines in space, otherwise you get osteoperosis and if you spend long enoug in space then returning to an atmospheric gravitational planet like earth or mars is a death sentance. Your body can't take it cause the changes from space cause permenant damage.
