First complete run unlocks a harder version of the map and one more boss who's way tougher than the final boss of the first run.
Basically. Tho there's various side quests, hidden paths and objectives that unlock other things. Hidden bosses as well. I'm not sure what are the conditions to getting some of them as when I've played online with randoms, more things seemed available or the randoms knew the secrets to unlocking other areas. Might've just been in their game objectives, as certain unlock objectives only show on the hosts side, I believe.
