Yes. There are differences GENETICALLY and PHYSICALLY. I posted a genetic study a few years back. In general, the Semitic speakers (like the Habesha) in East Africa have slightly more non-African ancestry than the Cushytic speakers (like the Oromo). Most likely this happened during the time when Ethiopia controlled Southern Arabia as a colony and there was probably some mixing between the Arabs and the Semitic Ethiopians.

The Semitic speaking Ethiopians tend to have slightly lighter skin and shorter physical stature. The Cushytic speaking Ethiopians have darker skin in general and have more of a elongated look like Africans who live in the plains (i.e. the Samburu and Maasai). Now obviously there has been mad mixing between these groups so you have many Oromos who look like Habesha and vice-versa. But in general based on what I've seen from watching YouTube videos of the different peoples in Ethiopia this is what you generally see.

I have no idea if there are cultural differences so I'll let the people who are actually Ethiopian answer that. I've only looked into the genetics and the physical features.
