The phone call was actually placed to Mission Control in Houston. Mission control connected it to the “Manned Space Flight Network” (MSFN) - which has dish antennas scattered around Earth so that at least one of them was facing towards the moon no matter the time of day or night. From there, the signal travel by radio the 240,000 miles to the Lunar Excursion Module (LEM), and because the crew were actually working outside during the call, the signal was sent out by a second radio in the LEM to antennas attached to the backpacks carried by Armstrong and Aldrin.

Their response came back via the same path, but in reverse.

Since the moon is about 1.3 seconds away from the Earth - their responses to Nixon were delayed by at least 2.6 seconds - probably much more because of the time from the MSFN dishes back to Mission Control and thence to Nixon and delays within the equipment used.

Many of the subsequent videos had those delays edited out…so it seems like an immediate conversation - when it really was not. However, they pretty much kept it to rehearsed statements rather than an actual conversation.
