Two spirits aren't actually a thing among indigenous tribes of America. That was made up. There were effeminate men that did women duties and the same were for masculine women. But they didn't have two spirits, they were what we call queer. If anything the indigenous people would say that the Spirit has no gender. But that's not true either.

Two sexes isn't a western idea. It's pretty universal. But because of biological deformities and religious/spiritual practices, other names were invented to describe these people.

The irony is that prior to be christianized they were totally accepting of LGBT members of their clans, people who fell under that umbrella were called Takatāpui. It was actually a common feature throughout Polynesian culture pre-colonization. Hawaii’s LGBT were called Aikāne and in Samoa they were called Fa'afafine.​

They are male, but the society they lived in dictated that they be castrated so they wouldn't reproduce, so they could work in the palace, or as punishment. They didn't stop being male just cause they had no balls.

These terms refer to categories of hijra functioning. Sex and gender do go together for sure. Biology influences behavior and women and men have very different types of behaviors. People with anomalies are called by different names and there are plenty of words for it, even in western terms. Metrosexuals, tomboys, queer.

Trans people has always existed. There were trans people in Africa who got snatched up in the slave trade. They will always be trans people no matter how hard you try to erase them
